1 полимеразная цепная реакция протяженных фрагментов
[греч. polymeres — состоящий из многих частей, многообразный; лат. re- — приставка, обозначающая повторность действия, и actio — действие; лат. fragmentum — обломок, отрывок]ПЦР (см. полимеразная цепная реакция), с помощью которой осуществляется синтез длинных фрагментов ДНК (до 5—10 тыс. п.н. и более). В этом случае для проведения реакции обычно используют термостабильные ДНК-полимеразы Pfu или Deep Vent (см. термостабильные ДНК-полимеразы); иногда реакцию проводят с двумя разными ДНК-полимеразами. В отличие от стандартного варианта ПЦР при проведении амплификации длинных фрагментов ДНК повышают рН реакционной смеси и включают в нее денатурирующие агенты.Толковый биотехнологический словарь.
